促红细胞生成素对兔慢性高眼压模型视网膜神经节细胞的保护作用 被引量:1

摘要 目的探讨促红细胞生成素(EPO)对兔慢性高眼压模型的视网膜神经节细胞的保护作用。方法新西兰大白兔35只,随机分为3组,Ⅰ组:正常对照组5只;Ⅱ组:治疗组即促红素玻璃体注射组15只;Ⅲ组:损伤组15只。Ⅱ组和Ⅲ组左眼前房注射卡波姆眼液制成慢性高眼压模型,schiotz眼压计测量眼压。Ⅱ组制模成功后开始向玻璃体腔注射r HuEPO3μl 3 000 U/ml,每周1次,连续3周。每组取10只眼球固定后行石蜡切片,做HE染色及TUNEL染色,检测各组实验结果。结果与Ⅰ组相比较,Ⅱ组和Ⅲ组模型眼的视网膜神经节细胞数明显减少,差异具有统计学意义(P<0.05);与Ⅲ组比较,Ⅱ组模型眼视网膜神经节细胞数多于Ⅲ组模型眼,差异具有统计学意义(P<0.05);TUNEL染色Ⅱ组和Ⅲ组模型眼视网膜神经节细胞凋亡数均高于对照眼,差异具有统计学意义(P<0.05);Ⅱ组模型眼TUNEL染色视网膜神经节细胞凋亡数少于Ⅲ组模型眼,差异具有统计学意义(P<0.05)。结论促红细胞生成素对兔慢性高眼压模型的视网膜神经节细胞有保护作用。 Objective To explore the neuroprotection of erythropoietin on rabbit retinal ganglion cells in chronic ocular hypertention model.Methods 35 rabbits without eye diseases were randomly divided into 3 groups:group Ⅰ:normal group(5 rabbits),group Ⅱ:erythropoietin treated group(15 rabbits),group Ⅲ:balanced salt solution treated group(15 rabbits).Rabbits in group Ⅱ and Ⅲ were induced with left eye by forcing pefusion of balanced salt solution into the anterior chamber as chronic ocular hypertenfion model.Right eyes were self-control eyes.No standard eyeballs were discarded and l0 eyeballs were selected and fixed in each group.(The retinal ganglion cells were detected by HE staining.The apoptosis cells were detected by TdT-dUDP terminal nick end-labeling(TUNEL) technique.Results The RGCs quantity of model eyes in group Ⅱ and Ⅲ were less than that of group Ⅰ and self-control eyes,there were significant diferences(P0.05).But the RGCs quantity of model eyes in group Ⅱ were more than that of group Ⅲ,there were significant diferences(P0.05).The TUNEL-positive cells of model eyes in group Ⅱ and Ⅲ were more than that of group Ⅰ and self-control eyes,there were significant diferences(P0.05).But the TUNEL-positive cells of model eyes in group II were less than that of group Ⅲ,there were significant diferences(P0.05).Condusion erythropoietin could provide neuroprotection for RGCs in rabbit chronic glaucoma model.
