摘要
目的探讨γ干扰素诱导蛋白10(IP-10)对呼吸道合胞病毒(RSV)感染所致小鼠肺部炎性病变的影响。方法 40只BALB/c小鼠随机分为4组,每组10只。对照组正常饲养,IP-10组单纯腹腔注射IP-10,RSV组只感染RSV,IP-10+RSV组腹腔注射IP-10后感染RSV。感染后第5 d,将小鼠处死,取肺脏,称重,计算小鼠肺指数,取左肺做病理分析及间质性肺炎病理评分。结果 1)肺指数:IP-10+RSV组为(1.99±0.25)%,与RSV组(1.51±0.16)%比较差异有统计学意义(P<0.01);2)病理变化:IP-10组与对照组均未见明显炎症反应,RSV组可见肺间质及血管周围炎性细胞浸润,IP-10+RSV组肺间质、支气管、血管处出现严重炎症反应;3)间质性肺炎病理评分:IP-10+RSV组为10.4±0.9,与单纯感染RSV组7.5±0.7比较差异有统计学意义(P<0.01)。结论 IP-10水平增高可加重RSV所致小鼠肺部炎性病变。
Objective To determine the role of IP-10 in pulmonary inflammation caused by respiratory syncytial virus.Methods Forty female BALB/c mice were divided into four groups.The control group was fed a normal diet,an IP-10 group was injected IP-10 alone,an RSV group was infected with RSV alone,and an IP-10+RSV group was infected with RSV after injection of IP-10.Mice were sacrificed on the fifth day after RSV infection and a lung was removed and weighed to calculate the lung index.The left lung was removed for pathological analysis and to calculate the interstitial pneumonia score.Results(1) Lung index: The IP-10+RSV group had a significantly higher(P0.01) index of(1.99±0.25)% in comparison to that of the RSV group(1.51±0.16)%.(2) Pathological analysis: The control group and the IP-10 group had no significant inflammation,the RSV group had moderate interstitial and perivascular infiltration,and the IP-10+RSV group had severe inflammation in the interstitial,peribronchial,and perivascular regions.(3) Interstitial pneumonia scores: The IP-10+RSV group had a significantly higher(P0.01) score of(10.4±0.9) in comparison to that of the RSV group(7.5±0.7).Conclusion An increased level of IP-10 may lead to more severe pulmonary inflammation caused by RSV.
