摘要
将实验室从陕北石油污染土壤中分离出来的S3#菌用牛肉膏液体培养基在摇床中以30℃、160 r/min恒温培养48 h,离心、用磷酸盐缓冲液淋洗3次后,用磷酸盐缓冲溶液配制成菌悬液。然后,用磷酸盐缓冲溶液将菌悬液稀释成不同倍数,以磷酸盐缓冲溶液作为空白对照,用WFJ7200可见分光光度计在600 nm下测量菌悬液的吸光度。得出结论,溶液稀释倍数和OD600是乘幂关系,关系式为y=1.402 5x-0.935 3,R2=0.998 8。
The S3# bacteria which is isolated from oil-contaminated soil in the northern Shaanxi is cultured with beef extract liquid as medium under the condition of shaker 30 ℃,160 r/min for 48 hours,centrifugation,washing with phosphate buffer for three times,and in the preparation of bacterial suspension by phosphate buffer.Then we dilute the bacterial suspension into different concentration multiples with the phosphate buffer,use phosphate buffer as the blank contrast,and the absorbance of bacterial suspension can be measured in 600 nm by using WFJ7200 Visible Spectrophotometers.The result is that the relationship between solution dilution multiples and OD600 is the power function,and the formula is y=1.402 5 x-0.935 3,R2 = 0.998 8.
