摘要
目的:建立HPLC测定维A酸在小鼠骨髓中含量的方法。方法:色谱柱为Diamonsil C18柱(150mm×4.6 mm,5μm),流动相为甲醇-水-冰醋酸(95∶5∶0.5),流速为1.0 mL·min^-1,检测波长为350 nm。外标法定量。结果:维A酸的线性范围为0.01-10μg·mL^-1(r=0.999 9),最低检测限为0.5 ng。日间与日内精密度均〈5%。低、中、高3个浓度的平均绝对回收率为74.5%,方法回收率为99.3%。结论:该方法灵敏度高、准确,可用于小鼠骨髓组织中维A酸浓度的测定。
Objective: To establish an HPLC method for determining all-trans retinoic acid(ATRA) concentration in mouse bone marrow.Methods: Shimadzu C18(150 mm×4.6 mm,5 μm) was applied;MeOH-H2O-HAc(95∶5∶0.5) was used as a mobile phase;flow rate was 1.0 mL·min^-1;and the detective wavelength was 350 nm.Results: The standard curve of ATRA was linear in the range of 0.01~10 μg·mL^-1(r=0.999 9).The limit of detection was 〈0.5 ng.The RSDs of intra-day and inter-day were 5%.The average extraction recovery was 74.5%,and the average recovery of the assay was 99.3%.Conclusion: The present study provides a sensitive and accurate method for the determination of ATRA in mouse bone marrow.
