摘要
本实验建立高效液相色谱法测定原料和复合预混料中低聚壳聚糖含量。采用ODSC18柱(150mm×4.6mm,5μm),以磷酸盐缓冲液(pH=2.8~3.0)-乙腈(95+5,V+V)为流动相,流速为1.5mL/min,检测波长为195nm,柱温为35℃。在此色谱条件下,低聚壳聚糖在0.056~0.450mg/mL浓度范围内呈良好线性关系,r=0.9998,方法平均回收率为98.8%,相对标准偏差(RSD)为1.1%(n=9)。本法操作简便、快速、结果准确,可作为低聚壳聚糖含量测定的质控方法。
A method for determination of low molecular weight chitosan in raw materials and compound premix by HPLC was developed.The separation was performed on ODSC18 column(150 mm×4.6 mm,5 μm)with mobile phase of phosphate buffer solution(pH=2.8 ~ 3.0)-acetonitrile(95+5,V+V) and flow rate was 1.5 mL/min.The determination was done at 35℃ for the column temperature and at 195 nm for the detection wavelength.Under above experimental conditions,the calibration curve for low molecular weight chitosan was linear in range of 0.056 ~ 0.450 mg/mL(r=0.9998).The average recovery was 96.1 %(n=9,RSD=1.1 %).The results obtained by experimental verification showed the method was simple,rapid and reliable for the determination of low molecular weight chitosan in raw materials and compound premix.
