响应面法优化肺炎克雷伯氏菌产酸性脲酶发酵培养基 被引量:1

Optimization of Fermentation Medium for Acidic Urease Producted by Klebsiella Pneumonia ssp using Response Surface Methodology

摘要 为了提高酸性脲酶的酶活力,设计响应面方法优化酸性脲酶产生菌肺炎克雷伯氏菌(Klebsiella pneumoniassp.)的发酵培养基。通过部分因子试验,从8种成分中筛选对酸性脲酶活性有显著影响的组分,再利用中心组合试验进一步优化培养基组成。试验结果表明,蛋白胨和KH2PO4对酸性脲酶活性有显著影响,优化后的培养基组成(g/L):葡萄糖8,酵母膏1.8,NaCl5,尿素8,Na2HPO40.9,MgSO40.2,蛋白胨6.015。在此条件下,模型预测发酵液中酸性脲酶酶活力可达28.74U/mL,比优化前酶活提高了67%。 In order to enhance activity of acidic urease,response surface methodology was used to optimize the fermentation medium of Klebsiella pneumonia ssp in this study.Firstly,fractional factorial design was adopted to evaluate the influence of 8 main factors.Then central composite design was used for further optimization.Results showed that concentrations of peptone and KH2PO4 had significant influence on acidic urease,and the optimized medium composition was as follows(g/L):glucose 8,yeast extract 1.8,NaCl 5,urea 8,Na2HPO4 0.9,MgSO4 0.2,peptone 6.015.Under the optimized culture condition,the activity of acidic urease was up to 28.74 U/mL,and 0.67 times higher than that in the original medium.
