MMP-2及TIMP-1、TIMP-2在自然流产绒毛和蜕膜组织中的表达及意义 被引量:7

Expression of matrix metalloproteinase-2 and tissue inhibitor of metalloproteinase-1,2 in chorionic villi and deciduas from women with spontaneous abortion

摘要 目的探讨基质金属蛋白酶-2(MMP-2)及金属蛋白酶组织抑制剂-1(TIMP-1)和金属蛋白酶组织抑制剂-2(TIMP-2)在早孕自然流产绒毛和蜕膜组织中的表达及意义。方法采用S-P免疫组织化学染色的方法,对31例自然流产者(实验组)及36例人工流产者(对照组)绒毛及蜕膜组织中MMP-2和TIMP-2的表达进行分析;采用逆转录聚合酶链反应(RT-PCR)技术对绒毛组织中MMP-2及TIMP-1的mRNA表达量进行分析。结果无论是绒毛组织还是蜕膜组织中MMP-2的表达两组之间均无统计学差异;但实验组两种组织中的TIMP-2强阳性(Ⅲ级)表达率均显著低于对照组(P<0.01);实验组TIMP-1 mRNA的表达量显著低于对照组(P<0.05)。结论自然流产绒毛和蜕膜组织中MMP-2的正常表达及TIMP-1、TIMP-2的低表达导致MMP-2与TIMP-1及TIMP-2的比值升高,该比值失调可能与自然流产发生有关。 Objective To investigate the expression of matrix metalloproteinase 2(MMP-2),tissue inhibitor of metalloproteinase 1(TIMP-1) and tissue inhibitor of metalloproteinase 2(TIMP-2) in chorionic villi and deciduas from spontaneous abortion women.Methods Immunohistochemical method(S-P) was employed to detect the expression of MMP-2 and TIMP-2 in chorionic villi and deciduas from women with spontaneous abortion(31 cases) and normal pregnant women(36 cases).Reverse transcriptase-ploymerase chain reaction(RT-PCR) was used to detect mRNA expression levels of MMP-2 and TIMP-1 in chorionic villi.Results There was no significant difference of MMP-2 expression in the two groups.While the strong positive(degree Ⅲ) expression rate of TIMP-2 in chorionic villi and deciduas from abortion group was significantly lower than that in the normal pregnant group(P0.01),and the mRNA expression level of TIMP-1 was significantly lower than that in the control group too(P0.05).Conclusions The higher ratio of MMP-2 to TIMP-1 or TIMP-2 in chorionic villi and deciduas,as a result of the normal expression of MMP-2 and the decreased expression of TIMP-1 and TIMP-2,may contribute to the spontaneous abortion.
