大黄素对人鼻咽癌裸鼠移植瘤的放射增敏作用研究 被引量:3

Enhancement Effect of Emodin on Radiosensitivity of Human Nasopharyngeal Carcinoma Transplanted in Nude Mice

摘要 目的评价广西产何首乌中大黄素对人鼻咽癌裸鼠移植瘤的放射增敏作用。方法通过皮下注射肿瘤细胞的方法建立人鼻咽癌裸鼠移植瘤模型,荷瘤鼠随机分成7组:空白对照组、溶媒组、大黄素组、单纯放射治疗组、大黄素联合放射治疗组,其中大黄素组和联合治疗组按照药物高、低剂量各分为两个亚组。经处理后各组隔日测量移植瘤的最大径,计算肿瘤生长延缓天数,利用增敏系数评价大黄素的放射增敏作用,HE染色,观察瘤组织病理学特征,计算核分裂率。结果单纯照射组肿瘤生长受到一定程度的抑制,和对照组相比,肿瘤生长延缓时间为4.28 d;高、低剂量大黄素联合放疗组较其他实验组肿瘤生长明显受到抑制,和单纯照射组相比,肿瘤生长延缓时间分别为6.90和6.77 d(P<0.05)。高、低剂量大黄素对鼻咽癌裸鼠移植瘤放射治疗的增敏值分别为1.612和1.601。高、低两个剂量大黄素联合放射治疗组的核分裂率分别是(3.80±0.45)%和(3.43±0.34)%,与对照组比较差异有统计学意义(P<0.05)。结论大黄素对人鼻咽癌裸鼠移植瘤具有放射增敏作用。 OBJECTIVE To evaluate the radiosensitivity of emodin isolated from Guangxi Polygonum multiflorum Thunb.on Human Nasopharyngeal Carcinoma Transplanted in nude mice.METHODS The model of carcinoma transplanted in nude mice was established via hypodermic injection of human nasopharyngeal carcinoma cell line(CNE-1).Mice bearing nasopharyngeal carcinoma were randomly divided into seven groups: control group,solvent group,emodin group,simple radiotherapy group,combination of emodin and radiotherapy group.The emodin group and combination group were divided into 2 subgroups according to the high dose and low dose of emodin.The maximum tumor diameters of every groups were measured every other day.The tumor growth delay was calculated.The enhancement factor(EF) was used to evaluate the radiosensitive enhancement.The pathological changes were observed by hematoxylin and eosin staining(HE staining) and the mitotic index(MI) was calculated.RESULTS Simple radiotherapy inhibited the tumor growth to a certain degree compared with control group,and the mean time of tumor growth delay was 4.28 d.The mean time of tumor growth delay were 6.90 d in high-dose and 6.77 d in low-dose,compared with the simple radiotherapy group respectively(P〈0.05).The EF values were 1.612 in high-dose group and 1.601 in low-dose group.The mitotic indexes(%) were(3.80±0.45) in high-dose group and(3.43±0.34) in low-dose group,respcetively.The mitotic index of combined groups were significantly lower than that in control group(P〈0.05).CONCLUSION Emodin can enhance the radiosensitivity of nasopharyngeal carcinoma-xenotransplantation.
