乳酸阈强度运动对糖尿病大鼠骨骼肌IRS-2蛋白含量及磷酸化的影响

摘要 观察乳酸阈强度运动对糖尿病大鼠骨骼肌胰岛素受体底物-2(IRS-2)蛋白含量及磷酸化的影响.在制备糖尿病大鼠模型后将SD大鼠分为3组:正常对照组(n=10),糖尿病对照组(n=10)和糖尿病运动组(20m/min组(n=10)).运动组每天运动50min,共6周.用Westernblot免疫印迹法检测骨骼肌细胞IRS-2蛋白磷酸化程度.结果显示:与糖尿病对照组相比,糖尿病运动组IRS-2蛋白含量升高24.4%(P<0.01)、磷酸化提高37.2%(P<0.01).本实验提示,乳酸阈强度运动对IRS-2蛋白含量和磷酸化程度产生明显影响. In this paper,we will explore effects of LT exercise on the protein content and tyrosine phosphorylation of IRS-2 in rats with diabetes.Thirty SD rats were divided into three groups:normal control group(NC,n=10),diabetes mellitus group(DMC,n=10)and exercise group(EXE,n=10)after DM model setting.The exercise groups did the running exercise each day for 50min with the intensity of 20m/min,lasting for six weeks.The content and tyrosine phosphorylation of IRS-2 protein were measured by Western bolt analysis.The results show that the content and tyrosine phosphory-lation of IRS-2 protein in exercise group enhanced up to 24.2%(P〈0.01)and 37.2%(P〈0.01)respectively,compared with the DMC group,but still keeping in a lower level compared with the NC group.The results hint that the LT exercise can affect the content and tyrosine phosphorylation of IRS-2 protein in skeletal muscle in DM rats.
