摘要
从盐腌皮上分离、筛选得到1株蜡状芽孢杆菌(Bacillus cereus SZ-4),该菌株经发酵后得到的粗酶液可在12 h内、无硫化物的条件下,将猪皮、牛皮、羊皮等完全脱毛。对该菌株发酵所产脱毛蛋白酶的酶性质进行研究,结果表明:脱毛蛋白酶的分子量约40 kDa,最适作用温度为60℃,最适作用pH为10.0,在40℃以上时热稳定性较差,K+、NH4+、Mg2+、Zn2+和Ca2+对脱毛蛋白酶有不同程度的激活作用,其中NH4+、Mg2+、Zn2+在一定浓度内具有明显的激活作用,Cu2+和Ba2+对脱毛蛋白酶有抑制作用,Na+在不同浓度范围对脱毛蛋白酶有不同的作用。
The crude enzymatic solution of a Bacillus cereus strain isolated from salt pickling skin could completely remove hairs of pigskin,cow leather and sheepskin without sulfide in 12 h.The results from studying enzymatic property of dehairing protease showed that the molecular weight of the dehairing protease was about 40kDa,the optimum reaction temperature and pH was 60℃ and 10.0,the thermal stability was poor above 40℃,K+、NH+4、Mg2+、Zn2+ and Ca2+ could activate dehairing protease to a certain extent,NH+4,Mg2+ and Zn2+ with a certain concentration could obviously activate dehairing protease,Cu2+ and Ba2+ had inhibitory effect on dehairing protease and Na+ with different concentration had different effect on dehairing protease.
