摘要
目的探讨美托洛尔对充血性心力衰竭大鼠血浆NE、AngⅡ的影响。方法实验选用清洁级雄性SD大鼠60只,随机分为正常对照组(n=10),模型组50只皮下注射异丙肾上腺素2次(170mg/kg),6周后应用超声心动图检测,以左室射血分数≤45%确定造模成功,将心力衰竭大鼠模型随机分为对照组、美托洛尔治疗组8mg/(kg.d),18周使用左心导管进行血流动力学检查,取血利用酶联免疫方法 (ELISA)检测NE和放免法检测血浆AngⅡ,并取心肌观察病理形态学特征。结果①与正常对照组相比,模型组心功能指标明显恶化,血浆NE、ANGⅡ显著升高,心室重量指数显著升高,心室重构明显;②与模型组比较,美托洛尔组心功能明显好转,血浆NE、ANGⅡ明显下降,心室重量指数明显降低,心室重构好转。结论美托洛尔能通过降低心力衰竭大鼠血浆NE、ANGⅡ的浓度,而改善心功能,逆转心室重构。
Objective To explore the effect of metoprolol on plasma Renin and Angiotensin Ⅱ in fats with heat failure.Methods 60 Male Sprague-Dawley rats were randomly allocated to the ISO-induced chronic heart falilure group(n=50) receiving two subcutaneous injections of 170mg ISO per-kilogram of body and the healthy normal group(NOR group,n= 10) receiving two subcutaneous injections of 0.25ml normal saline.At 6th week,the left ventricular ejection fraction(LVEF) was measured by echocardiogram,the remaining rats,with an left ventricular ejection fraction≤45%,were randomly divided into two groups: control group(n=10),metoprolol group 8 mg/(kg·d)(n=10).the drugs were given i.g.At 18th week,then the hemody namic parameters were detected,including heart rate,left ventricular end diastolic pressure,LV systolic pressure;LVWI was measured after LV morphopathological examination,the Renin and Angiotensin Ⅱ in plasma were detected by ELISA and radio-immunity method,the mass ratio of heart ventricle and left ventricle to body was measured. Results ①Compared with the NOR group,the HR、LVEDP,LVWI and the Renin and Angiotensin Ⅱ in plasma were significantly increased(P0.01).But LVSP and ±dp/dtmax were significantly decreased in the CON group.②Compared with the CON group,the HR、LVEDP、LVWI and the Renin and AngiotensinⅡin plasma were decreased distinctly.the LVSP and±dp/dtmax were increased distinctly(P0.01) in the MET group.Conclusion Metoprolol can inhibit the Renin and AngiotensinⅡin plasma,inprove heart function and reverse ventricular remodeling with congestive heart failure.
