分泌型卷曲相关蛋白基因启动子甲基化状态检测在急性白血病中的意义 被引量:5

The significance of methylation status of secreted frizzled-related protein gene promoter in acute leukemia

摘要 目的 探讨分泌型卷曲相关蛋白(SFRP)家族基因启动子异常甲基化状态与急性白血病(AL)发生发展的关系.方法 采用甲基化特异性PCR(MSP)检测AL患者、AL细胞系和正常人外周血单个核细胞中SFBP(1、2、4、5)基因启动子区的甲基化状态.结果 正常人单个核细胞中不存在SFRP基因的甲基化.AL患者中,SFRP1、2、4、5基因甲基化总发生率分别为35.6%(31/87)、25.3%(22/87)、12.6%(11/87)、17.2%(15/87),其中在急性髓系白血病(AML)患者中的甲基化率分别为33.9%(20/59)、23.7%(14/59)、6.8%(4/59)、10.2%(6/59),在急性淋巴细胞白血病(ALL)患者中的甲基化率分别为39.3%(11/28)、28.6%(8/28)、25.0%(7/28)、32.1%(9/28).SFRP1、2、5基因在HL60、NB4、Molt-4和Jurkat细胞系中均呈完全甲基化状态,SFRP4在NB4、Molt-4和Jurkat细胞系中呈完全甲基化状态,在HL60细胞系中则呈部分甲基化状态.结论 在AL患者及细胞系中,SFRP(1、2、4、5)基因已出现高频率甲基化.推测SFRP基因的异常甲基化模式与AL的发生密切相关,其可能成为诊断和监测AL的基因标志物. Objective To investigate the relationship between promoter hypermethylation of secreted frizzled-related protein (SFRP) gene and acute leukemia (AL). Methods We examined the promoter methylation starus of SFRP1, 2, 4 and 5 in primary or relapsed AL patients, cell lines ( HL60,NB4, Molt-4 and Jurkat) and peripheral blood mononuclear cells from healthy people with methylationspecific PCR (MSP). Results None of the normal mononuclear cells showed methylation of any SFRP genes. The frequencies of aberrant methylation among the samples were 33.9% (20/59) for SFRP1,23.7%(14/59) for SFRP2, 6. 8% (4/59) for SFRP4 and 10. 2% (6/59) for SFRP5 in acute myelocytic leukemia (AML), and 39.3% (11/28) for SFRP1, 28.6% (8/28) for SFRP2, 25.0% (7/28) for SFRP4 and 32. 1% (9/28) for SFRP5 in acute lymphoblastic leukemia (ALL). Hypermethylation of SFRP1, 2, 5genes were present in the 4 AL cell lines. SFRP4 was methylated in NB4, Molt-4 and Jurkat cell lines.However, methylation and unmethylation of SFRP4 were both detected in HL60. Conclusions Hypermethylation of SFRP genes is a common event in the evolution of AL. Methylation of SFRP genes might serve as potential independent biomarkers for early detection of AL.
