摘要
运用高速逆流色谱法(High-speed counter-current chromatography,HSCCC)分离纯化丹酚酸B。建立了石油醚-乙酸乙酯-甲醇-水-甲酸两相溶剂系统,确定最佳纯化工艺条件为:转速1800 r.min-1,流速1.0 mL.min-1,温度30℃。纯化后的丹酚酸B经HPLC检测,纯度达98.3%。表明高速逆流色谱法分离纯化丹酚酸B具有快速、简便、纯度高等优点,可用于丹酚酸B的制备。
The separation and purification of salvianolic acid B by HSCCC were studied.A solvent system petroleum ether-acetic ether-methanol-water-formic acid was selected as solvent.The optimum pruification conditions were determined as follows:rotative velocity 1800 r·min^-1,flow rate 1.0 mL·min^-1,temperature 30℃.Analyzed by HPLC,the purity of pruified salvianolic acid B was 98.3%.The results showed that purification of salvianolic acid B by HSCCC was quick,convenient with high purity and yield.It could be used in salvianolic acid B preparation.
