肾性高血压大鼠模型制作的改良方法 被引量:10

摘要 目的探讨新型钛合金夹经大鼠背部切口进行双侧肾动脉夹闭制作肾性高血压模型的可行性。方法选用94只雄性Sprague-Dawley(SD)大鼠,随机分为两组:实验组74只采用经背部开口,行双侧肾动脉夹闭术;对照组20只仅游离双侧肾动脉不予狭窄,余同实验组。术后6周测定成活率、成模率、血压和心脏质量指数。结果术后成活率、成模率高,术后6周模型组血压均在160 mmHg以上。两组血压和左心室质量指数有显著性差异(P<0.05)。结论经背部开口行大鼠双侧肾动脉夹闭术是一种安全性高和手术操作简便的新方法。 Objective To raise an optimized method for renovascular hypertensive model by reforming two-kidney two-clip method with new titanium alloy clips via back bi-incising in rats. Methods 94 male SD rats were randomly divided into two groups.In experimental group,both renal arteries of 74 rats were occluded by clips from back bi-incising.Both renal arteries in 20 control rats were liberated but not narrowed,other operation was as same as model rats.After 6 weeks,the survival rate,the model rate,blood pressure and the ratio of LVW/BW were calculated. Results The model rats had high survival rate after operation.Compared with control rats,the blood pressure and the ratio of LVW/BW in model rats were significantly increased.The blood pressure at 6 weeks after operation was more than 160 mmHg in model rats. Conclusion The optimized method of renovascular hypertensive model by narrowing both renal arteries from back bi-incising in rats is a simple operating and safe approach,suggesting the practical utility of this new method.
