摘要
目的探讨云南白药对四氯化碳所致肝纤维化的影响及量效关系。方法在CCl4所致小白鼠肝纤维化动物模型中,分别应用小剂量和大剂量云南白药水剂治疗,观察肝组织脯氨酸羟化酶活性、脯氨酸及羟脯氨酸含量和肝细胞变性、坏死、胶原纤维增生等指标的变化,图像分析测定肝组织胶原纤维相对面积。结果小剂量组、大剂量组分别与正常组及肝纤维化对照组比较,脯氨酸羟化酶活性、脯氨酸及羟脯氨酸含量、胶原纤维相对面积、肝细胞变性、坏死、胶原纤维增生等指标差异均有统计学意义(P<0.05或0.01);小剂量组与大剂量组比较,肝组织学改变无明显差异(P>0.05),其余指标间差异均非常显著(P<0.01)。结论云南白药对四氯化碳所致肝纤维化具有明显的保护作用,并有较明显的量效关系。
Objective To explore the effect of Yunnan baiyao on rat liver fibrosis induced by carbon tetrachloride and the relationship between dosage and effect.Methods Fibrotic rats models were established by administration of carbon tetrachloride.Small and large dosage of Yunnan baiyao(SYTG and LYTG) were administered to them intragastrically using an oral catheter.The activity of prolyl-hydroxylase,the content of proline and hydroxyproline were detected,the relative area of collagen was measured by image analysis and the change of pathology was observed by light microscope.Results The side-effect of Yunnan baiyao was not found in treated group.Compared with normal control group(NCG) and liver fibrosis control group(LFCG),the activity of prolyl-hudroxlase,the content of proline and hydroxyproline,the area of collagen and the change of pathology had significant differences in small and large treat groups(P0.05 or 0.01).The difference was not significant in the change of pathology(P0.05) between SYTG and LYTG,but significant in the other indexes.Conclusion Yunan baiyao has significant protective effect on liver fibrosis induced by carbon tetrachloride and decreases the degree of rat liver fibrosis.The effect of large dosage is better than that of small dosage.
