摘要
目的:探讨大鼠大脑海马立体定向注射β-淀粉样蛋白1-40(Aβ1-40)对caspase-3蛋白表达的影响及通络救脑口服液的干预作用。方法:140只SD大鼠随机分为空白对照组、假手术组、模型组、盐酸多奈哌齐组、通络救脑口服液低、中、高剂量组(12、24、48mg.kg-1.d-1)共7组。采用大脑海马立体定向注射凝聚态Aβ1-40诱导老年性痴呆动物模型,药物干预4周后,电镜观察海马组织超微结构的变化,实时免疫印迹法检测caspase-3蛋白的表达。结果:电镜观察结果显示:模型组大鼠海马神经元变性,通络救脑口服液能改善大鼠海马神经元病理改变。免疫印迹结果显示:与假手术组比较,模型组大鼠海马组织caspase-3蛋白表达显著升高(P<0.05);与模型组比较,通络救脑口服液组(12、24、48mg.kg-1.d-1)大鼠海马组织caspase-3蛋白表达均显著减少(P<0.05)。结论:海马注射Aβ1-40后大鼠海马组织caspase-3蛋白表达水平明显增高,而通络救脑口服液可以抑制海马组织促凋亡基因caspase-3蛋白的表达,从而发挥其抗老年性痴呆神经细胞凋亡的作用。
Objective:To observe the effect of TLJN Oral Solution on protein caspase-3 expression in brain tissue of amyloid-β1-40 injection into brain induced experimental AD rats in vivo.Methods:140 SD rats were enrolled in the study,and they were equally randomly divided into blank controls group,sham controls group,negtive control group,positive control group and experiment group(12mg?kg-1?d-1,24mg?kg-1?d-1,48mg?kg-1?d-1),A rat model of AD induced by amyloid-β1-40 sterotaxis injection the bilateral Hippocampus.All rats were sacrificed at 5 week after treatment for 4 weeks.Change of hippocampus is observed by electron microscope.Protein caspase-3 expression in brain tissue was assayed by immunoblotting test.Results:Hippocampus is found denatured in model rats,and TLJN Oral Solution can alleviate the damage of hippocampus neurons.The protein caspase-3 expression was up-regulate expression in model groups compared with the sham group(P0.05).In experiment groups the protein caspase-3 expression was down-regulate expression compared with the model group(P0.05).Conclusions:amyloid-β1-40 injection the bilateral Hippocampus can lead to the over-regulate expression of protein caspase-3 in hippocampus tissue.TLJN Oral Solution may down-regulate expression of protein caspase-3.
