摘要
以矮紫杉的愈伤组织及腋芽为繁殖体,对人工种子的制作工艺、胚乳配方和萌发条件等进行初步研究。采用滴注法在海藻酸钠凝胶系统中添加不同种类及浓度的可溶性淀粉、诱导子和植物激素制备人工种子。结果表明,3.0%海藻酸钠加2.5%CaCl2为人工种子的最佳种皮配方;添加1.5%的可溶性淀粉能提高人工胚乳的保水性,24℃下储藏15 d之后失水率为25.29%;在人工胚乳中添加诱导子,以10-6mg/L水杨酸效果最好,种子颗粒均一透明;在人工胚乳中添加NAA能提高人工种子的萌发率,以1.5 mg/L NAA的效果最好,培养30 d后萌发率达38%。
Callus and axillary buds originated from Taxus cuspidata CV. Nana were used as propagule to produce artificial seeds, and the prepare procedures, optimization of artificial endosperm and germination conditions were investigated. The results showed that the combination of 3.0% sodium alginate with 2.5% CaCl2 was the best combination for artificial seed coat, and 1.5% soluble starch could elevate the artifical endosperm with higher water-retention capacity compared with that of 3.0 % sodium alginate alone, and the loss of moisture was only 25.29% after 15 d of storage at 24 ℃. The 10^-6 mg/L salicylic acid is the better for its uniform and transparent particles. Addition of 1.5 mg/L NAA can increase the germination rate of artifical seeds to 38% after 30 d of culture.
