摘要
利用海水双层平板法分离获得蛭弧菌LBd02-1,从基因水平上进行鉴定,并对其生物学特性进行了初步研究。研究结果表明,蛭弧菌LBd02-1对嗜水气单胞菌、鳗弧菌、大肠杆菌和霍乱弧菌等革兰氏阴性菌有较好的裂解作用;对金黄色葡萄球菌、枯草芽孢杆菌等革兰氏阳性菌的裂解能力较弱。以鳗弧菌为宿主菌,探索了不同条件(温度、盐度、Ca2+、Mg2+)下蛭弧菌LBd02-1对鳗弧菌的裂解效果。试验结果表明,蛭弧菌LBd02-1在温度为25~30℃.盐度为10~30时,对鳗弧菌的裂解效果较佳;在Ca2+浓度为5~10 mmol/L、Mg2+浓度为0.5~2 mmol/L时,蛭弧菌LBd02-1的裂解能力也较好。
Bdellovibrio sp.LBd02-1 was isolated in the double-layer agar plate from sea mud and identified on genic lever.The biological characteristics of LBd02-1 provided the information for biological control of aquatic animal diseases.The results revealed that the G-bacteria(such as Aeromonas hydrophilia,Vibrio anguillarum,Esherichia coli and V.cholerae)were more beneficial to be lysed than the G+ bacteria(such as Saccharomyces cerevisial and Bacillus subtilis)by LBd02-1.Strain LBd02-1 can lyse the host bacteria Vibrio anguillarum easily in the temperature from 25 ℃ to 30 ℃ and the salinity from 10 to 30.We designed Ca2+ and Mg2+ concentration as 0,0.5,2.0,5.0,and 10.0 mmol/L,and found that the lysis characters were better as Ca2+ levels increased from 5 mmol/L to 10 mmol/L and Mg2+ from 0.5 mmol/L to 2 mmol/L.The results of the trial established the foundation to the further study,and made a reference to the production of microbial ecological agents.
