摘要
以野生重瓣棣棠花茎段为外植体,进行组织培养技术体系的建立。结果表明,启动培养时,外源激素为6-BA1.0mg/L+NAA0.1mg/L的效果最好,启动率达到93.33%;适合继代增殖的培养基是6-BA1.5mg/L+NAA0.1mg/L,增殖系数达到7.8;而1/2MS+6-BA1.0mg/L+NAA0.1mg/L+AC5g/L的培养基生根效果高于其他处理,生根率达到78.60%。
In this paper,the steam of Kerria japonica(Linn)DC.f.pleniflora(Witte)Rehd was used as explants to carry out its tissue culture.The results revealed that the best initiation rate of 93.33%was obtained after adding exogenous hormone of 1.0mg/L 6-BA and 0.1 mg/L NAA in medium.The multiplication coefficient of subculture in the most suitable subculture multiplication medium containing 1.5 mg/L 6-BA and 0.1 mg/L NAA could reach 7.8.The highest rooting rate of 78.60%was observed in the medium containing 1/2 MS,1.0 mg/L 6-BA,0.1 mg/L NAA and 5 g/L AC.
