摘要
目的:研究脑舒胶囊对阿尔茨海默病模型大鼠的防治效果及其作用机理。方法:双侧脑室注射Aβ25-35复制阿尔茨海默病大鼠模型;Morris水迷宫观察大鼠学习记忆能力;免疫组织化学法(SABC法)检测大鼠海马Bcl-2、Bax及caspase-3的表达;HE染色光镜观察海马神经元损伤情况。结果:与假手术组比较,模型组大鼠的逃避潜伏期明显延长,跨越虚拟平台的次数及在平台象限停留的时间明显减少;Bcl-2和Bax表达均有所增加,其平均光密度比值(Bcl-2/Bax)明显下降;Caspase-3阳性表达明显增高。与模型组比较,0.72mg/kg和1.44mg/kg脑舒胶囊可缩短大鼠逃避潜伏期,增加大鼠跨越虚拟平台的次数及在平台象限停留的时间;使Bcl-2阳性表达明显所增加,Bax阳性表达明显减少,升高Bcl-2/Bax值;明显降低Caspase-3的阳性表达;光镜观察发现模型大鼠海马神经元排列较紊乱、数量减少、体积变小,胞浆浓缩,胞核固缩深染,结构不清,各剂量的脑舒胶囊均能在一定程度上改善上述神经元损伤。结论:脑舒胶囊可改善阿尔茨海默病大鼠的记忆能力及Aβ25-35所致海马神经元损伤,其原因与其提高抗凋亡基因Bcl-2的表达,抑制促凋亡基因Bax表达和caspase-3表达,进而抑制海马神经元的凋亡有关。
Objective:To investigate the preventive and therapeutic effect of Naoshu Capsule on AD rats and the mechanism.Methods:AD model was induced by injecting Aβ25-35 in the bilateral lateral ventricles of rat,learning and memory capability of rats was observed with Morris water maze,expression of Bcl-2,Bax and caspases-3 in hippocampus was detected by SABC,hippocampal neuron injuries was observed by HE staining.Results:Compare with sham operation group,the rats in model group show longer escape latency,and less times to cross the virtual platform and staying on the platform,the expression of Bcl-2 and Bax strengthens,the ratio of their optical density(Bcl-2/Bax) obviously decreased,the positive expression of Caspase-3 obviously enhanced.Compare with the model group,in the 0.72mg/kg and 1.44mg/kg Naoshu Capsule group,there is shorter escape latency of rats,more times to cross the virtual platform and longer tine to stay on the platform,strenthen the positive expression of Bcl-2,inhibit the positive expression of Bax,increase the ratio of their optical density(Bcl-2/Bax) ;decrease the positive expression of Caspase-3;and in the light microscope,the hippocampus neurons in the model group have irregular arrange ment,samll number,small volume,thick plasm,pycnotic and thick nucleus,blur structure;the improvement occur in all the different Naoshu Capsule Group.Conclusion:Naoshu Capsule can improve AD rats'memory amd alleviate neurons'damage by Aβ25-35.The mechanism may be that the Naoshu Capsule increased the expression of Bcl-2,inhibited the expression of Bax and caspase-3,finally inhibit the apotisis of hippocampus neurons.
