摘要
目的:通过观察不同浓度硒制剂对连续厌氧培养的变形链球菌(S.mutans)生长及产酸情况的影响,观察硒对变形链球菌的作用。方法:将复苏的变形链球菌标准株(Ingbritt C),分别等量接种于含不同浓度硒的TS液体培养基中。采用厌氧菌连续培养技术,通过分光光度计检测菌液OD值,绘制生长曲线,并通过pH仪检测菌液pH值;培养24h终止后,接种于平皿进行菌落(CFU)计数。结果:实验组变形链球菌的生长曲线在生长对数期较对照组低(培养第2~8h),硒浓度0.8ppm组菌液OD值低于其他组(P〈0.05);各实验组与阴性对照组菌液pH值无明显差别(P〉0.05);硒浓度为0.8ppm组菌落计数低于其他实验组与阴性对照组(P〈0.05)。结论:一定浓度的硒(0.8ppm)可使变链菌生长受到抑制。值得注意的是,在该组变链菌数目明显减少的情况下,细菌培养液的pH值与其他组无显著性差异,其原因尚待进一步研究。
Objective:To observe the effect of Selenite on the growth and glycolysis of S.mutans.To find the minimal inhibitory concentration of selenite on the growth of S.mutans.Methods:Different concentrations of selenite were separately added to TS medium as experimental group.S.Mutans(Ingbritt C) grew in the TS medium with different concentrations of Selenium.The concentration of selenite in negative control group was zero,and the microbe was absent in blank control group.All cultures were incubated at 37℃ anaerobically in an atmosphere of 80% of nitrogen(v/v),10 % of hydrogen(v/v) and 10% of carbon dioxide(v/v) for 24 hours.The OD values of culture medium were measured by using a spectrometer at 2、3、4、5、6、8、12、16、20、24 hours to describe the growth curve of the S.Mutans,and also the pH values of the medium were measured by using pH electrode.The colony forming units(CFU) were obtained by inoculate the dilute cultures at 24 hours in media.Results:The sigmoid curve of the S.Mutans growth at 0.8ppm concentration of Selenium was suppressed than that in the other group.The OD value of culture medium was also lower than that in the other group at exponential phase(2~8h),but there was no significant difference at plateau phase.There was no significant difference of pH value between experimental groups and control group.Conclusion: This experiment indicates the trace element Selenium has inhibination effects on the growth of S.mutans at 0.8ppm concentration.However,there is no significant difference on the pH value among these groups while the growth of S.mutans being inhibinated.It still needs further study.
