摘要
目的观察壮骨镇痛胶囊对高转移人乳腺癌MDA-MB-231细胞抑癌基因nm23-H1和原癌基因c-myc表达的影响,探讨壮骨镇痛胶囊抑制MDA-MB-231细胞增殖和迁移的可能作用机制。方法采用RT-PCR法和免疫细胞化学法分别检测不同浓度壮骨镇痛胶囊含药血浆对MDA-MB-231细胞nm23-H1和c-myc基因mRNA及其蛋白表达的影响,并分析其对nm23-H1/c-myc蛋白比值的影响。结果 5%壮骨镇痛胶囊含药血浆显著抑制了MDA-MB-231细胞nm23-H1和c-myc基因的转录和翻译;1.25%壮骨镇痛胶囊含药血浆抑制了nm23-H1的转录和翻译以及c-myc基因的翻译,对c-myc基因的转录无显著影响;0.63%壮骨镇痛胶囊含药血浆对nm23-H1和c-myc基因的转录和翻译均无明显影响;5%和1.25%壮骨镇痛胶囊含药血浆显著提高了MDA-MB-231细胞nm23-H1/c-myc蛋白比值,0.63%壮骨镇痛胶囊含药血浆对nm23-H1/c-myc蛋白比值无显著影响。结论不同浓度壮骨镇痛胶囊含药血浆对乳腺癌MDA-MB-231细胞nm23-H1和c-myc基因的表达具有不同的作用,该药可能通过降低癌细胞c-myc基因表达继而提高细胞中nm23-H1/c-myc蛋白比值来降低癌细胞的增殖和迁移能力。
Objective In order to provide evidence of mechanisms of Zhuanggu Zhentong capsules,we investigated the impacts of Zhuanggu Zhentong capsules on the expressions of nm23-H1 gene and c-myc gene of breast cancer cell lines MDA-MB-231 in vitro.Methods The mRNA levels of nm23-H1 gene and c-myc gene were examined by reverse transcriptase PCR(RT-PCR).The nm23-H1 and c-myc proteins were assayed by immunocytochemistry.The ratio of nm23-H1/c-myc proteins was also analyzed.Results The transcriptions and translations of nm23-H1 and c-myc genes of MDA-MB-231 cells treated with 5% Zhuanggu Zhentong capsules contained plasma were obviously inhibited.There were obvious inhibitions on the transcriptions and translations of nm23-H1 gene and translations of c-myc gene in MDA-MB-231 cells after 1.25% Zhuanggu Zhentong capsules contained plasma treatment,but without obvious inhibition on c-myc gene expression.There were no obvious inhibitions on both the transcriptions and translations of nm23-H1 gene and c-myc gene in MDA-MB-231 cells after 0.63% Zhuanggu Zhentong capsules contained plasma treatment.The ratios of nm23-H1/c-myc proteins in MDA-MB-231 cells after 5% and 1.25% Zhuanggu Zhentong capsules contained plasma treatment were markedly improved,but there were no obvious effects on the ratio of nm23-H1/c-myc proteins after 0.63% Zhuanggu Zhentong capsules contained plasma treatment.Conclusion The inhibitive effects of Zhuanggu Zhentong capsules on the expression of nm23-H1 and c-myc genes in MDA-MB-231 cells are shown in dose-dependent manner.The inhibitive effects on proliferations and migrations of Zhuanggu Zhentong capsules may be related with the improvement of the nm23-H1/c-myc proteins ratio.
