摘要
采用流式细胞术方法测定多疣壁虎(Gekko japonicus)DNA含量(C值)。结果表明,多疣壁虎的C值为2.928 pg.C-1,计算其基因组大小约为2.86×109bp,雌性和雄性壁虎的C值分别为2.934 pg和2.923 pg。数据统计经独立样本t检验性别间无显著差异。相同方法测定了另外两种爬行动物北草晰(Takydromusseptentrionalis)和原尾壁虎(Hemidactylus bowringii)的C值,分别是(2.10±0.03)pg.C-1和(2.04±0.03)pg.C-1,并计算其基因组大小,北草晰为2.054×109bp,原尾壁虎为1.995×109bp。
The present study aims to investigate the DNA content(C-value) of Gekko japonicus by flow cytometry.The result showed that the C-value of Gekko japonicus is 2.928 pg·C-1 with the calculated genome size approximately of 2.86×109 bp.The C-values for female and male are 2.934 pg and 2.923 pg,respectively.Independent-samples T test indicated that the C-values of different genders have no significant difference.Meantime,we observed the C-values of two other reptile species,and found that C-values of Takydromus septentrionalis and Hemidactylus bowringii are(2.10±0.03) pg·C-1 and(2.04±0.03) pg·C-1.By this means the calculated approximate genome size of Takvdromus septentrionalis is 2.054×109 bp and that of Hemidactylus bowringii is 1.995×109 bp.
