摘要
目的观察低压缺氧对兔动脉粥样硬化斑块胶原含量的影响,并对其机制进行初步探讨。方法日本大耳兔随机分为常氧对照组和低压缺氧组两组,高胆固醇饮食12周建立兔动脉粥样硬化(atherosclerosis,AS)病变模型后,取主动脉内膜组织分别采用Woessner法、western blot法和实时荧光定量逆转录聚合酶链反应(RT-PCR)法检测斑块组织中胶原含量、人核转录因子(NF-κB)蛋白、肿瘤坏死因子(TNF-α)、白细胞介素6逆转核糖核酸(IL-6mRNA)表达。结果低压缺氧能明显降低斑块中胶原蛋白含量,提高NF-κB蛋白、TNF-α和IL-6mRNA表达(P<0.05)。结论低压缺氧能明显降低斑块中胶原蛋白含量,其机制可能与促进炎性反应,升高NF-κB蛋白、TNF-α及IL-6mRNA表达有关。
Objective To study the effects of hypobaric hypoxia on collagen content in atherosclerotic plaques of rabbit models and to initially investigate its mechanism.Methods Rabbit atherosclerotic models were set up by high-cholesterol diet.The contents of NF-κB and collagen in plaque were detected by Western blot and woessner method,respectively.Expression levels of TNF-α,IL-6 mRNA were determined with real-time PCR.Results In hypobaric hypoxia group,collagen in atherosclerotic plaques was decreased markedly,TNF-α,IL-6 mRNA and NF-κB protein found in atherosclerotic plaques were elevated.Conclusion Hypobaric hypoxia could decrease the collagen content in rabbit atherosclerotic plaques,which may be concerned with its promoting inflammatory action.
