摘要 目的探讨胃蛋白酶水解黄粉虫蛋白制取复合氨基酸液的工艺。方法以黄粉虫为原料,以碱法提取黄粉虫蛋白,采用胃蛋白酶水解黄粉虫蛋白制取复合氨基酸液。通过单因素试验探讨了底物浓度、pH值、酶浓度、水解温度及水解时间与水解率的关系,采用正交试验L16(45)优选水解工艺条件。结果最佳酶解工艺条件为:底物浓度为1∶5,pH值4.0,酶浓度1.5%,水解温度60℃,水解时间5 h。水解率达到24.15%。结论按最佳条件水解完毕后,通过酶灭活,中和,脱色等处理可得到黄色澄清的黄粉虫蛋白复合氨基酸液。所制得的复合氨基酸种类齐全,必需氨基酸含量高。 Aim To explore the technology for composite amino acids from Tenebrio molitor protein by pepsin.Methods Alkaline was used to extract proteins from Tenebrio molitor.The relationships among substrate concentration,pH,enzyme concentration,hydrolysis temperature and time,and degree of hydrolysis were analysed through single factor test by preparing multI-amino acids with pepsin hydrolysis.As the same time,the optimal condition of hydrolysis was discussed by orthogonal test L16(4^5).Results The optimal enzyme hydrolysis condition is determined as follows:substrate concentration was 1∶5,pH 4.0,enzyme concentration 1.5%,hydrolysis temperature 60℃,hydrolysis time 5 h,and the hydrolysis rate 24.15%.Conclusion The full range of amino acids,including high content of essential amino acid,was obtained by the optimal hydrolysis condition.
