摘要
为寻找与结缕草(Zoysia japonica steud.)抗寒性状相关的数量性状位点(QTLs),利用生态条件具明显差异的2个日本结缕草品系室兰(Zoysia japonsic cv. Muroran)和俵山北(Zoysia japonsic cv. Tawarayama Kita)及其假测交产生的F1代86个个体为材料,研究其在人工低温胁迫条件下叶片的半致死温度(LT50)及可溶性糖、可溶性蛋白含量及过氧化物歧化酶(SOD)活性的变化,并以447个SSR标记构建的日本结缕草遗传连锁图谱为基础,对抗寒相关的性状可溶性糖、可溶性蛋白含量及SOD活性进行了QTL定位分析。结果表明:F1群体的可溶性蛋白含量与叶片LT50呈显著负相关(P〈0.05),可溶性糖含量及SOD活性与叶片LT50呈极显著负相关(P〈0.01)。分别定位到与叶片可溶性糖、可溶性蛋白含量和SOD活性相关的QTL各1个,分布于3个连锁群上,QTL的LOD值介于2.19~2.42之间,单个QTL可解释性状表型变异的范围在13.3%~13.8%。
To find quantitative trait loci(QTLs) relating to cold resistance of Zoysia japonica,two different ecotypes(Muroran and Tawarayama Kita) and a pseudo-testcross F1 population(86 individuals derived by crossing 'Zoysia japonica cv.Muroran' with 'Zoysia japonica cv.Tawarayama Kita') were used for mapping and QTL analysis.A genetic linkage map with 447 SSR markers was constructed for zoysiagrass(Zoysia japonica Steud.).The semi lethal temperature(LT50),soluble protein,soluble sugar content and SOD activity of leaves of F1 population were analyzed.Results indicate a significant negative correlation between the leaf LT50 and the soluble protein content of F1 population(P〈0.05).Also very significant negative correlations were observed between the leaf LT50 and the soluble sugar content as well as the SOD activity of F1 population(P〈0.01) under the treatment of 0℃ for 2 days.Three QTLs,located on tenth,thirteenth and seventeenth linkage group respectively,relating to the soluble protein,the soluble sugar content and the SOD activity were detected.The score of LOD ranged from 2.19 to 2.42,and the phenotypic variation explained by individual QTL varied from 13.3% to 13.8%.
