磺胺喹恶啉人工抗原合成及鉴定 被引量:2

Synthesis and identification of sulfaquinoxaline antificial antigen

摘要 国内首次用重氮化法将磺胺喹恶啉(SQ)分别与牛血清白蛋白(BSA)和卵清白蛋白(OVA)偶联,制备免疫抗原SQ-BSA和包被抗原SQ-OVA。紫外扫描法与SDS-PAGE凝胶电泳法鉴定表明偶联成功,偶联比分别为10.1∶1、9.8∶1。将SQ-BSA免疫BALB/C小鼠,获得了高效价、敏感性好的抗SQ血清,表明ELISA方法鉴定偶联成功,为进一步制备抗SQ单抗奠定了基础。 The diazotization was adopted to link SQ to the carrier proteins BSA and OVA to prepare the immunogen SQ-BSA and test coating antigen SQ-OVA.Also, SQ-BSA and SQ-OVA were confirmed by ultraviolet scanning and SDS-PAGE.The conjugationratio of SQ-BSA and SQ-OVA were 10.1:1 and 9.8:1, respectively.The polyclonal antibody with high titre and sensitivity were also gained by immunizing the BALB/C mice with SQ- BSA.These results indicated that the hapten SQ was successfully linked to the carrier proteins,which also lay the foundation for further gain monoclonal anti-SQ.
