摘要
目的观察甘遂醇提物及不同极性溶剂萃取物对体外培养MCF-7、A549、HepG2细胞生长的影响。方法采用甘遂醇提物及其环己烷、乙酸乙酯的萃取物和水溶性组分,分别与体外培养的MCF-7、A549、HepG2细胞作用48h,采用噻唑蓝(MTT)法测定各供试品对细胞增殖的影响。结果甘遂醇提物对MCF-7有较好的抑制作用,半数抑制浓度(IC50)为202μg·mL-1;甘遂乙酸乙酯萃取物和环己烷萃取物对MCF-7有较好的抑制作用,IC50分别为552和378μg·mL-1,仅在高浓度时对A549和HepG2细胞增殖有抑制作用;甘遂水溶性组分高浓度时对A549细胞和HepG2细胞增殖有促进作用。结论甘遂抑瘤成分可能存在于乙酸乙酯、环己烷萃取部位,促肿瘤生长成分可能存在于水溶性组分。
Objective To investigate the growth-modulating effects of the extracts and different polar parts of Kansui on the proliferation of MCF-7,A549,HepG2 cells.Methods The extracts of Kansui,acetate extract,cyclohexane extract and water extract from Kansui were co-cultured with MCF-7,A549,and HepG2 cell lines,and the cell proliferation was evaluated by MTT colorimetric assay.Results The ethanol extract of Kansui obviously reduced MCF-7 proliferation,with IC50 values as 202 μg·mL-1;Acetate extract and cyclohexane extract from Kansui suppressed MCF-7 as well,with IC50 as 552 μg·mL-1 and 378 μg·mL-1,respectively.They stimulated the growth of A549 and HepG2 cells at a high concentration.The water soluble parts from Kansui stimulated A549 and HepG2 cells proliferation at high levels.Conclusion The anticancer components in Kansui could be exist in acetate extracts and cyclohexane extracts,while the cancer-stimulating compotents be in the water extracts from Kansui.
