摘要
目的:建立同时测定血清中7种抗抑郁类药物和5种抗精神病类药物浓度的液相色谱-串联质谱(HPLC-MS/MS)方法。方法:血清样品中加入还阳酚(内标,西地那非中间产物),采用乙腈直接沉淀法处理样品。分析柱为XbridgeTMPhenyl柱(150mm×2.1mm,5μm);流动相为含0.02%甲酸水溶液-含0.02%甲酸乙腈溶液(35:65),等度洗脱,流速为0.3mL·min-1,进样量为20μL。采用电喷雾离子源,多离子反应监测(MRM),正离子扫描进行测定。结果:7种抗抑郁类药物和5种抗精神病类药物线性范围在2~1000μg·L-1,线性相关系数均大于0.9931,各浓度的批内和批间RSD〈10%,提取回收率为102%~112%,方法准确度为89.3%~107.9%。结论:本方法操作简便,特异性强,灵敏度高,可同时进行7种抗抑郁类药物和5种抗精神病类药物的血药浓度监测。
Objective:To establish an HPLC-MS/MS method for determination of seven antidepressants and five antipsychotics in human serum.Methods:The serum sample was precipitated with acetonitrile after the addition of internal standard(huanyangfen).The chromatographic separation was performed on XbridgeTM Phenyl column(150 mm×2.1 mm,5 μm) with the mobile phase of 0.02% formic acid water-0.02% formic acid acetonitrile(35:65) at a flow rate of 0.3 mL·min-1.The injection volume was 20 μL.The deprotonated ion of analyte was ionized by turbo ion spray and was detected in positive ionization by multiple reaction monitoring mode(MRM).Results:The linear concentration ranges of the calibration curves for seven antidepressants and five antipsychotics were 2-1000 μg·L-1.The coefficients of linearity were more than 0.993 1.RSDs of withinbatch and between-batch were less than 10%.The average recovery ranged from 102% to 112%.The accuracy ranged from 89.3% to 107.9%.Conclusion:The method is simple and quick with high specificity and sensitivity,and it is suitable for quantitative determination of seven antidepressants and five antipsychotics in human serum.
