慢性阻塞性肺疾病大鼠骨骼肌组织20s蛋白酶体C_2亚基的基因及蛋白表达的研究 被引量:7

摘要 目的探讨慢性阻塞性肺疾病(COPD)大鼠骨骼肌组织内蛋白降解的信号分子——泛素-蛋白酶体系统中20s蛋白酶体C2亚基的基因和蛋白表达水平及意义。方法采用单纯熏香烟法复制COPD大鼠动物模型。反转录-聚合酶链式反应(RT-PCR)技术检测伸趾长肌内20s蛋白酶体C2亚基的mRNA表达水平;蛋白免疫印迹(Westernblot)法测定大鼠伸趾长肌内20s蛋白酶体C2亚基的蛋白表达。结果 RT-PCR结果显示,伸趾长肌组织中20s蛋白酶体C2亚基的mRNA表达(IOD值)较对照组增加,差异有统计学意义(P<0.01);Westernblot结果显示,伸趾长肌组织中20s蛋白酶体C2亚基的蛋白表达较对照组增强,差异有统计学意义(P<0.01)。结论 COPD大鼠骨骼肌泛素-蛋白酶体系统中20s蛋白酶体C2亚基的基因和蛋白表达均显著增强,提示骨骼肌内泛素-蛋白酶体途径活性上调,是蛋白降解增强的原因之一。 Objective To study the expression of mRNA and protein of 20s proteasome subunit C2 of ubiquitin-proteasome pathway in skeletal muscle in COPD rats.Methods COPD rat models were copied by simple smoking method.Reverse transcription-polymerase chain reaction(RT-PCR) was used to determine mRNA expression of 20s proteasome subunit C2 in extensor digitorum longus(EDL),and Western blot used to detect its protein expression.Results The mRNA expression of 20s proteasome subunit C2 in EDL(IOD value)increased as compared with control group(P〈0.01),and its protein expression was higher than that of control group,the difference was significant(P〈0.01).Conclusion The expressions of mRNA and protein of 20s proteasome subunit C2 of ubiquitin-proteasome pathway in skeletal muscle increase significantly in COPD rats,suggesting that the activity increase of ubiquitin-proteasome pathway leads to an increase of protein degradation.
