脑红蛋白与Caspase-3在大鼠海马CA1区的相关性 被引量:2

摘要 目的通过对脑缺血再灌注后大鼠脑红蛋白(NGB)与Caspase-3的测定及其在时间表达上的相关性,分析NGB的抗凋亡作用。方法将60只健康SD大鼠随机分为实验组和对照组。实验组采用Pu lsinelli-Brierley’s四血管阻塞法建立急性全脑缺血再灌注损伤模型。应用免疫组化和原位杂交法检测缺血15 m in再灌注后3、6、12、24、48 h Caspase-3和NGB的表达变化。HE染色存活神经元。结果实验组大脑海马CA1区存活锥体细胞数随缺血时间延长逐渐减少。实验组Caspase-3 mRNA及蛋白在缺血再灌注后3 h开始表达,24 h达高峰;6、12、24、48 h与对照组比较有统计学意义(P<0.05);实验组NGB mRNA及蛋白在缺血再灌注后3、6 h弥散表达,12 h后随时间延长表达逐渐减少,与对照组比较差异有统计学意义(P<0.05),Caspase-3与NGB呈负相关性。结论 NGB阻止神经细胞的凋亡,且与Caspase-3呈负相关关系。
