镰刀菌产纤维素酶液体发酵工艺研究 被引量:1

摘要 以镰刀菌为对象,对其产羧甲基纤维素酶(CMCase)的液体发酵工艺进行研究,结果表明:稻草秸秆粉末为最佳碳源,NaNO3为最佳氮源,且碳源∶氮源(C∶N)为6∶1时酶活力最大;不同的无机盐对产酶有一定的影响,AlCl3具有一定的促进作用,而CuSO4.5H2O和AgNO3则有明显的抑制作用;表面活性剂吐温-80在低浓度(0.04%)下对产酶有促进作用。同时考察了装液量、发酵液初始pH、接种量、菌龄、发酵温度及发酵时间对产酶的影响,确定适宜的培养条件为:75 mL/250 mL,初始pH7,接种量10%,菌龄42 h,发酵温度及发酵时间分别为30℃和96 h。通过正交试验确定的最佳发酵条件为:接种量9%,菌龄42 h,发酵时间108 h,发酵温度32℃。 Fermentation procedure for the cellulase were established in liquid by Fusarium.sp in the current work.The results showed that rice straw powder was the best carbon source and NaNO3 was the best nitrogen source.When the ratio of C/N was 6∶1,the enzyme yield was highest.AlCl3 played a role in promoting the yield,however CuSO4·5H2O and AgNO3 significantly demonstrated a negative effect.Low concentration(0.04%) of tween-80 had a promoting effect on enzyme yield.Also,the effects of some factors,including liquid volume in shaking flasks,initial pH,inoculum volume,inoculum age,fermentation temperature and fermentation time on the enzyme yield were further investigated,and the appropriate culture conditions were 75mL/250mL,pH 7,10% inoculum volume,42-hour-old inoculum,30℃ for 96 h,respectively.By orthogonal experiment,the optimal fermentation condition was established as the 9% inoculum amount,42-hour-old inoculum,32℃ fermentation for 108h.
