摘要
目的探讨γ-分泌酶抑制剂DAPT对人宫颈癌Hela细胞系增殖和凋亡的影响。方法体外培养人宫颈癌Hela细胞;MTT法检测不同浓度DAPT(1.0、5.0、10.0μmol/L)对Hela细胞增殖的抑制作用;流式细胞术分析不同浓度DAPT对Hela细胞凋亡率及细胞周期分布的影响。结果 3种浓度的DAPT均能显著抑制Hela细胞的增殖,抑制率分别为22.847%、32.492%、46.565%,作用呈明显剂量依赖性;不同浓度的DAPT能使Hela细胞凋亡率明显增加,分别为,且使S期、G2/M期细胞比例减少,G0/G1期细胞比例显著增加,作用呈剂量依赖性。结论 DAPT对体外培养的人宫颈癌Hela细胞具有抑制增殖、调节细胞周期、诱导凋亡的作用。
Objective To investigate the effect of DAPT on the proliferation and apoptosis in human cervical carcinoma cell line Hela.Methods Human cervical carcinoma cell line Hela was cultured in vitro system.To study the inhibiting effect of DAPT on the proliferation of Hela cells by MTT assay.Flow cytometry analysis technique were used to determine the changes of cell cycle distribution and cell apoptosis proportion after the treatment with different concentrations of DAPT.Results Three different concentrations of DAPT could obviously inhibit the proliferation of Hela cells.The inhibition ratios were 22.847%,32.492% and 46.565%.Apoptosis analysis showed that DAPT increased the cell apoptosis proportion(8.462%,13.683%,18.690%) and the rate of cells in G0/G1 phase of Hela cells,while decreased the rate of cells in S and G2/M phase.Conclusions DAPT can inhibit the in vitro proliferation of human cervical cancer Hela cells,adjust cell cycle distribution and accelerate cell apoptosis.
