灯盏细辛提取物对痴呆大鼠学习记忆能力和胆碱酯酶活性的影响 被引量:6

摘要 目的探讨灯盏细辛提取物对痴呆大鼠学习记忆能力及胆碱酯酶活性的影响。方法 Wistar大鼠42只,随机分为5组;用双侧脑室注射β淀粉样蛋白联合腹腔注射D-半乳糖方法建立痴呆大鼠模型。采用Morris水迷宫方法检测大鼠学习记忆能力、比色法检测乙酰胆碱酯酶(AChE)和丁酰胆碱酯酶(BuChE)活性。结果与正常组和假手术组相比,模型组大鼠在定向航行和空间探索中逃避潜伏期时间和穿越平台时间延长,穿越平台次数减少(P<0.05);脑组织中AChE和BuChE活性升高,血浆中AChE和BuChE活性降低(P<0.05),经过灯盏细辛治疗后痴呆大鼠学习记忆能力有所提高,胆碱酯酶活性趋向于正常(P<0.05)。结论灯盏细辛能够提高痴呆大鼠学习记忆能力,促进胆碱酯酶活性趋向于正常。 Objective To investigate the influence of Herba Erigenomtis( HE) extract on learning and memory ability and cholinesterase activity in rats with dementia. Methods Wistar rats were randomly divided into 5 groups. The animal model with dementia was produced by bilateral intracterthroventricular injection with β-amyloid peptide ( Aβ) 25-35 and abdominal cavity injection with D-galactose. Learning and memory ability of rats was detected by Morris Water Maze test,and the activities of acetycholinesterase (AChE) and butyrylcholinesterase (BuChE) were measured by Ellman's colorimetric method. Results In the rats with dementia in the model group as compared with normal and sham operation groups,the escaping latent period and the time in passing the platform were obviously increased ( P 0. 05) ,and the times of crossing the platform significantly were declined ( P 0. 05) ; the activities of AChE and BuChE were increased in brain tissues and decreased in plasma ( P 0. 05) . After treatment by HE extract,the ability of learning and memory was improved in the rats with dementia and the activity of cholinesterase in brain tissues and plasma was shown a tendency to recover to normal level ( P 0. 05) . Conclusion HE extract may improve the learning and memory ability of the rats with dementia and show a tendency to recover the activity of cholinesterase to normal level.
