摘要
[目的]初步建立农村稳定塘底微生物结构分析的PCR-DGGE分子生物学分析技术,并对我国南北方2个地区农村稳定塘底泥中的微生物群落进行简要分析。[方法]提取杭州、西安两地郊区农村稳定塘底泥系统内微生物的总DNA,对其16SrDNA的V3可变区进行PCR扩增,并进行变性梯度凝胶电泳(DGGE),利用软件对DGGE图像进行聚类分析。[结果]两地农村稳定塘底泥中细菌种类较为丰富,有明显而相似的优势种群。[结论]南北两地区农村稳定塘底泥微生物多样性基本相似,无明显变化。
[ Objective ] PCR-DGGE technology was established to analyze the bacterial community of rural stabilization pond, and two bacterial communities of rural stabilization pond in north and south China were analyzed briefly. [ Method] Bacterial genomic DNAs were extracted di- rectly from the rural stabilization pond bottom activated sludge which was took from Xi'an and Hangzhou. V3 region of the 16S rDNA genes were amplified, and separated by DGGE, then DGGE patterns were analyzed by associated softwares. [ Result ] Species in rural stabilization ponds of the two areas were rich and similar. [ Conclusion] The bacterial diversity was abundant and relatively stable in rural stabilization pond in north and south China.
