摘要
目的探讨异体异位移植睾丸中生精细胞退化的可能机制。方法以新生1~2 d小鼠睾丸组织为供体,免疫缺陷的雄性去势裸鼠为受体进行组织移植,分别在移植后不同时间(3 d和1~8周共9个时间段)收集移植物组织,提取移植物总RNA,采用半定量RT-PCR检测不同发育阶段移植物中凋亡相关基因fas、bcl-2、bax和caspase3以及DNA损伤修复基因xrcc1、ogg1、ercc1、brca1和brca2的表达,并与相应年龄段正常小鼠睾丸组织中的基因表达进行比较分析。结果与正常发育的小鼠睾丸组织相比,小鼠睾丸移植物中生精细胞退化增加,凋亡相关基因bcl-2表达降低,fas、bax和caspase3表达增加;DNA损伤修复基因xrcc1、ogg1、ercc1、brca1和brca2的表达量均有不同程度的降低。结论新生小鼠睾丸组织移植到免疫缺陷裸鼠体内后,生精细胞退化增加,这可能与促凋亡基因表达增加和抑制凋亡基因表达降低,以及DNA损伤修复基因表达降低有关。
Objective To investigate the expression levels of apoptosis-related gene and DNA repair gene in ectopic grafted mouse testes using the animal model. Methods Testicular tissue grafting was performed in castrated male immunodeficient mice with testes from neonatal Kun-ming mice aging 1~2 days. Grafts were collected at differ- ent time intervals (3 days and 1~8 weeks, respectively). The expressions of apoptosis-related genes fas, bcl-2, bax and caspase3 as well as DNA repair genes xrccl, oggl, erccl, brcal and brca2 in grafts were detected by RT-PCR Simultaneously, the expressions of the above genes in normal Kun-ming mice with relevant ages were also detected Results The expression of bcl-2 decreased and the expressions of fas, bax and caspase3 were increased in the obtained grafts at different experimental intervals compared with those in testes of normal Kun-ming mice, whereas the expres- sions of xrcc 1, oggl, ercc 1, brcal and brca2 were all decreased in the obtained grafts at different experimental intervals Conclusion Germ cells were degenerated in grafts of immunodeficient mice, which might be associated with the upregulated expressions of pro-apoptotic genes and the downregulated expressions of anti-apoptotic genes and DNA repair genes
