摘要
目的:研究1号、2号三胶扶正合剂在小鼠体内对放化疗所致的骨髓抑制和免疫功能低下的拮抗作用。方法:腹腔注射环磷酰胺(CTX)复制低白细胞小鼠模型,皮下注射CTX复制免疫抑制小鼠模型,分别以复方阿胶浆为阳性对照药,设立1号、2号三胶扶正合剂高、中、低给药剂量,测定外周白细胞数、骨髓有核细胞数、脾集落形成单位(CFU-S)及碳粒廓清能力,考察各组疗效及最佳用药剂量;小鼠灌胃给药考察急性毒性作用。结果:1号、2号三胶扶正合剂及复方阿胶浆均有一定的提升外周白细胞、促进骨髓有核细胞增殖及CFU-S增加,并增强免疫功能低下小鼠的网状内皮系统吞噬功能的作用,且1号合剂效果最好;1号、2号三胶扶正合剂及盐酸左旋咪唑片对小鼠网状内皮系统功能的增强作用无明显差异;小鼠的最大耐受量为150g·kg-1。结论:三胶扶正合剂能够拮抗放化疗所致小鼠的骨髓抑制和免疫功能低下,且安全无毒副作用,可为临床推广奠定理论基础。
OBJECTIVE:To observe the antagonistic effect of Ⅰ,Ⅱ Sanjiao fuzheng mixture against myelosuppression and low immune function induced by radiotherapy and chemotherapy in mice.METHODS:Low white blood cell and immune suppression model mice were established by intraperitoneal injection of CTX and hypodermic injection of CTX respectively using Compound A-jiao oral liquid,levamisole hydrochloride tablets(LMS)as positive control.High-dose,medium-dose and low-doseⅠ,ⅡSanjiao fuzheng mixture were set up to determine the peripheral leukocyte count,bone marrow nucleated cells,spleen colony-forming units(CFU-S)and carbon clearance ability.Therapeutic efficacy and optimal dosage of each group were investigated.Acute toxicity was studied after mice received drugs via i.g.gtt.RESULTS:Ⅰ,ⅡSanjiao fuzheng mixture and Compound A-jiao oral liquid had the functions of upgrading peripheral leukocytes,and promoting the value of bone marrow nucleated cells and CFU-S,and enhancing phagocytic function of reticuloendothelial system in mice with low immune function.ⅠSanjiao fuzheng mixture showed the best efficacy;no significant difference was observed in the improvement of reticuloendothelial system function betweenⅠ,ⅡSanjiao fuzheng mixture and levamisole hydrochloride tablets;MTD of mice was 150 g·kg -1.CONCLUSION:Sanjiao fuzheng mixture can be used to inhibit radiotherapy and chemotherapy-induced myelosuppression and low immune function.It is safe and non-toxic side effects.The study lays the theoretical foundation for clinical popularity.
