摘要
以华中农业大学康思农实验蜂场油菜蜂蜜为材料,研究了XAD-2树脂对油菜蜂蜜中黄酮类物质的柱层析分离条件。在考察4种不同因素对分离效果影响的基础上,得出的最佳分离条件是:蜂蜜上样量与树脂量的比为1.2:1;上样液pH=2;90%的甲醇为洗脱溶剂,并采用3倍柱床体积的甲醇量(洗脱速度为4.5ml/min),其加标回收率为82.8%。对柱层析分离得到蜂蜜黄酮样品采用LC/MS/MS(液质联用)分析,发现康思农油菜蜂蜜样品中含有槲皮素、异鼠李素、山萘酚、木樨草素、白杨黄酮、高良姜黄素、短叶松素、芹菜素、5,7-二羟基双氢黄酮和汉黄芩素等10种黄酮类物质。
The rape honey from the Kang Sinong bee field of Huazhong Agriculture University was used to study column chromatography conditions of XAD-2 resin about extracting flavonoids from honey. After considering four different factors impacting on the separation, the optimalizing conditions were: the ratio between sample quantity of honey and resin quantity, 1.2:1; pH of solution, 2; eluting solution,90% methanol, and the amount of volume of methanol, 3 times column bed (elution rate 4.5ml/min), and the recovery was 82.8%. After LC/MS/MS analyzing, Flavonoids isolated by XAD-2 resin in honey were: quereetin, isorhamnetin, kaempferol, luteolin, chrysin, galangin, pinobanksin, apigenin, 5,7-dihydroxy-flavanone and wogonin
