成纤维细胞激活蛋白P和转化生长因子在子宫内膜癌组织中的表达及其意义 被引量:8

摘要 目的探讨成纤维细胞激活蛋白P(FAP)和转化生长因子(TGFβ1)在正常子宫内膜和子宫内膜癌组织中的表达,及其与子宫内膜癌临床病理参数之间的关系。方法采用免疫组织化学链霉素抗生物素蛋白-过氧化物酶(SP)法对10例正常子宫内膜组织和54例子宫内膜癌组织中FAP和TGFβ1的表达情况进行了检测,分析其在正常子宫内膜组织中的表达规律及其与子宫内膜癌临床病理特征的关系。结果在54例子宫内膜癌组织的间质中FAP表达阳性率为88.89%(48例),而子宫内膜癌细胞少量表达,在正常子宫内膜组织中无表达。FAP表达与子宫内膜癌组织的侵袭程度有关、即与临床分期有关,与病理分级和淋巴结转移无关。提示FAP的表达与子宫内膜癌恶性表型有关、即随着子宫内膜癌恶性程度的增强FAP表达逐渐增多。在54例子宫内膜癌组织中TGFβ1表达阳性率为87.04%。TGFβ1表达与子宫内膜癌的临床分期和病理分级有关,与淋巴结转移无关。提示TGFβ1随着子宫内膜癌恶性程度的增加表达有不同程度的增强。FAP表达阳性组中TGFβ1阳性表达率(91.67%)高于FAP表达阴性组(50.00%),两者的表达呈正相关。结论子宫内膜癌细胞表达的TGFβ1可能诱导癌间质中FAP的表达,从而促进了肿瘤的演进。 Objective To explore the expression of Fibroblast Activation Protein and TGFβ1 in normal endometriums and endometrial carcinomas,evaluate clini-cal significance of FAP and TGFβ1.Methods Streptavidin-biotin peroxidase(SP) immunohistochemical technique was used to detect the expression of FAP and TGFβ1 in 10 cases of normal endometriums and 54 cases of en-dometrial carcinoma,respectively·The expression of FAP and TGFβ1 and the relationship between FAP and TGFβ1 and clinicopathological characteristics of endometrial carcinomawere analyzed.Results The results showed that FAP was detected in the stroma of 88.89%(48 in 58) of endometrial carcinoma while normal endometrium tissue was negative,and the expression of FAP in carcinoma cells was at a low level.FAP had a positive correlation with clinical stage of endometrial carcinoma,but not with pathological grades and lymph node metastasis.The rate of positive expression of TGFβ1 in endometrial carcinoma cells was 87.04%,and the expression of TGFβ1 in cervical carcinoma was correlated with clinical stage and pathological grades,but not with lymph node metastasis,the expression of FAP and TGFβ1 also had positive correlation in endometrial carcinoma.Conclusion TGFβ1 expressed in carcinoma cells may induce the expression of FAP in reactive stroma and then promote tumor progression.
