摘要
目的:研究水红花子对部分肝切除小鼠骨髓细胞增殖的影响。方法:参照文献方法制备小鼠2/3肝切除模型。给药7d后处死动物,摘取肝脏,计算肝指数;分离小鼠骨髓细胞,流式细胞仪检测DNA含量和细胞周期分布。结果:水红花子3.6、1.8 g/kg使肝切除小鼠骨髓细胞DNA含量降低,水红花子3.6、1.8、0.9 g/kg使模型鼠G0/G1期细胞数量增加,而G2/M和S期细胞减少,具有剂量依赖性。PH后小鼠的体重增加缓慢,术后7d时模型鼠的体重和残肝重量明显低于正常对照组。水红花子给药组小鼠的体重和残肝重量明显低于正常对照组,但和模型组比较没有显著性差异。结论:水红花子抑制肝切除小鼠骨髓细胞的增殖。
Objective:To explore the effect of fructus Polygoni orentalis(水红花子) on DNA content and cell cycle of bone marrow cells in partially hepatectomized(PH) mice.Methods:Healthy and PH mice which were subject to PH were divided into 5 groups: NS group,control group and fructus Polygoni orentalis groups,respectively,then were treated ig with 0.9% NaCl,and fructus Polygoni orentalis 3.6,1.8,0.9 g/kg daily,respectively.Sacrifice mice and remove the livers for calculating index of liver,bone marrow cells were isolated and detected by flow cytometer(FCM) after 7 days administration.Results:DNA content was decreased by fructus Polygoni orentalis(P〈0.01),the proportion of G0/G1 phase were increased,and G2/M and S phase were decreased(P〈0.01).Body weight(BW) of mice added slowly after PH 7 days,and the weight of residual liver lower than that of NS group.BW of fructus polygoni orentalis 0.9 g/kg obviously decreased,and index of liver of fructus Polygoni orentalis 3.6 g/kg male mice higher than that of control group(p〈0.05).Conclusion: Fructus Polygoni orentalis could inhibit the proliferation of bone marrow cells in PH mice.
