摘要
全长cDNA的获得是基因克隆的重要内容,也是基因组研究的重要内容之一。cDNA末端快速扩增技术(RACE)是获得全长cDNA的主要辅助手段之一,本文对RACE技术的原理、改进和近几年发展起来的新型RACE(包括RNA连接酶介导的RACE、oligo-capping等)作了综述。同时,对用干全长CDNA克隆的其他技术,如引物载体法、固相支持物法作了介绍。
Acquision of full-length cDNA is important both to gene cloning, and to genomicreseajrching. Rapid amplification of cDNA end (RACE) is one of research ways in this field.Principle' modification and recent-developed techniques (including RNA ligase mediated RACE'oligo-capping method etc) of RACE is summarized in the paper. Other useful methods to clonefull-length cDNA is also reported, such as vector primer method, solid support method, etc.
