补肾活血方调控Wnt/β-catenin信号通路保护关节软骨的机制研究 被引量:18

Study on mechanism of protective effect on articular cartilage by Kidney-tonifying and Blood Circulation-promoting Recipes through regulating Wnt/β-catenin signaling pathway

摘要 目的:探讨补肾活血方对实验大鼠骨关节炎关节软骨保护的机制。方法:以清洁级健康雌性SD大鼠为动物模型,分为Sham组、模型组和中药组。通过切除双侧卵巢并切断右侧膝交叉韧带建立骨关节炎动物模型,术后第4周开始灌服补肾活血中药,模型组、Sham组灌服等量的生理盐水,分别于术后8、12、16周取材,采用骨关节炎软骨病理变化评价系统(OARSI)评价关节软骨的病变,采用激光共聚焦显微镜下观察软骨细胞凋亡情况,运用免疫组化方法评估关节软骨中Wnt2、β-catenin、GSK3的含量,运用Western blot法检测GSK3β蛋白磷酸化水平。结果:术后8、12、16周,模型组较Sham组OARSI评分差异有显著意义(P<0.01),中药组较模型组OARSI评分差异有显著意义(P<0.05)。术后16周,模型组见大量软骨细胞凋亡,而中药组、Sham组见少许软骨细胞凋亡,中药组、Sham组与模型组比较,差异有统计学意义(P<0.05)。Wnt2、β-catenin、GSK3β在模型组中表达明显增强,补肾活血中药能下调Wnt2、β-catenin、GSK3β的表达。模型组GSK3β蛋白质磷酸化水平较低,中药组GSK3β蛋白质磷酸化水平较高。结论:补肾活血方通过调控Wnt/β-catenin信号通路保护关节软骨,其机制可能是通过保护或修复软骨细胞的完整性,重塑细胞的衰老平衡稳态,延缓软骨病损,或增强软骨修复能力,促进关节康复,可作为治疗骨关节炎有潜力的药物。 Objective:To evaluate the mechanism of protective effect on articular cartilage by Kidney-tonifying and Blood Circulation-promoting Recipes through regulating Wnt/β-catenin signaling pathway.Methods:30 SD rats were divided into sham group,model group and Chinese medicine group.The osteoarthritis models were obtained by bilateral ovariectomy and right cruciate ligaments of knee cutting off.Chinese medicine group were given Kidney-tonifying and Blood Circulation-promoting Recipes on the forth week after the operation,the model group and sham group were given the equivalent physiological saline,and the specimens were excised after 8,12,16 weeks.Taking the OARSI to evaluate the pathological changes of the articular cartilage,using confocal laser scanning microscope(CLSM)to observe chondrocyte apoptosis,using immunohistochemistry to measure the content of Wnt2,β-catenin,GSK3βin articular cartilage,using Western blot to measure activity change of GSK3β.Results: The OARSI scores were significantly higher in the model group than that in the sham group(P0.01),and as were significantly higher in Chinese medicine group than that in the model group(P0.05).The percentage of positive apoptotic chondrocytes in the model group was higher than that of Chinese medicine group and the sham group.The immunohistochemistry demonstrated that in model group Wnt2,β-catenin,GSK3βincreased,Chinese medicine group diminished.There was remarkable difference in the expression of GSK3βbetween the model group and Chinese medicine group(P0.05).Conclusion:Kidney-tonifying and Blood Circulation-promoting Recipes can treat osteoarthritis through regulating Wnt/β-catenin signaling pathway.
