摘要
目的:在硫酸脱氢表雄酮-多囊卵巢综合征(DHEA-PCOS)大鼠模型的基础上,加用人绒毛膜促性腺激素(HCG)和高脂饲料,建立一种新的改良多囊卵巢综合征-胰岛素抵抗(PCOS-IR)大鼠模型。方法:将52只21日龄SPF级雌性SD大鼠随机分为5组,按所设计实验方案造模,观察该模型体质量、脏器湿重、内分泌激素及代谢指标、组织形态学与超微结构的改变。结果:T+HCG组模型具有腹部生殖器周围脂肪积聚,高雄激素血症、高胰岛素血症、高胆固醇血症等内分泌、代谢异常改变,以及排卵抑制、卵巢不排卵及多囊改变更典型的PCOS特征。结论:改良PCOS-IR大鼠模型更接近PCOS临床病理生理,可为肥胖型PCOS模型的建立奠定研究基础。
Objective:The study is based on the PCOS rat model induced by Dehydroeplandrosterone(DHEA),and the rats are feed with Human Chorionic Gonadotropin(HCG)and High fat diet in order to establish the improved Puberty PCOS- IR rate model.Method:52 simples 21st day SPF SD female rats which weight controlled between 35-45g were randomized into five groups.According to the experimentation,to investigate the effect on weight,organ weight,hormonal,metabolized and morphology changes in the ovary of PCOS rat model.Result:The characteristics of T+HCG model group is:the rats had the abdominal obese--in particular fat pad which around reproductive organ obvious increased,and retained the high androgen syndrome as the endocrine characteristics.Simultaneously they had sugar fat metabolism unusual,such as the insulin resistance,the redemptive high insulin blood sickness as well as hypercholesterolemia.The ovary increased obviously,Ovarian cortex accumulated,the number in ovary pouch expansion graafian follicle were remarkable increased,ovulation suppression.Conclusion:The improved Puberty PCOS-IR rate model is closer to the PCOS pathology of human.It may be the foundation of the study of porky PCOS model.
