摘要
SOD属酸性蛋白酶,对pH、热和蛋白酶水解等特性比一般酶稳定。Cu/Zn—SOD,Mn—SOD,Fe—SOD三类SOD分子结构均为二聚体或四聚体,其活性中心都含有金属离子,金属离子的存在与否直接影响酶活,CN-可以使Cu/Zn-SOD失活,而Mn—SOD和Fe—SOD具有抗CN-能力。SOD作为药用酶使用存在半衰期短、分子量大,抗原性,易被蛋白酶水解等问题,因此,对SOD分子进行改造就显得十分重要。对SOD进行化学修饰,可以明显地提高SOD的稳定性。修饰后的SOD不仅完全保留了天然酶的活性,而且在耐热、耐酸、耐碱和抗胃蛋白酶水解能力方面都明显地优于天然SOD。鉴于SOD是一类重要的氧自由基清除剂,具有抗炎、抗衰老和抗辐射的作用,作为一类新型的治疗酶制剂已广泛应用在医疗、保健、食品等方面。
SOD, an acid protease, has more stable properties to pH, heat and so on. The molecular structures ofCu/Zn--SOD, Mn--SOD and Fe-SOD are all tetramer or dimer. There are metal ions in the activity centers of thethree kinds of SODs, which affects the activities of enzyme. CN-can make Cu/Zn-SOD lose it activities, whileMn--SOD and Fe-SOD both are resistant to CN-. As a kind of medical enzyme, SOD has the problems such as ashorter half-life, heavier molecular weight, zymogen resistance and being easy to be hydrolpsized by protease. Therefore, it becomes very important to reform the SOD molecular. Chemically embellishing SOD can obviously improveitS stability. The embellished SOD not only keeps all the activity of natural enzyme, but also bornes obviously superior to natural enzyme in resistant to heat, acid, alkaline and the ability of resisting pepsin hydrolysis. Due toresistant to inflammation, aging and radiation, SOD, as a new type of medical ferment preparation, has been widelyapplied to medical treatments, health care, food and so on.
