摘要
采用水提法从胡枝子中提取多糖,通过单因素和正交试验确定提取胡枝子多糖的最佳工艺。同时还对胡枝子多糖的抑菌、抗氧化、抗凝血药理活性进行了研究与分析。结果表明:料液比1:20(g/mL),提取温度100℃,提取时间60min,提取次数2次,为水提法提取胡枝子多糖的最佳工艺,最高提取率为3.75%。胡枝子多糖具有明显的抗菌活性。其中对大肠杆菌和四联球菌的抑菌活性比较强,且浓度越高作用越强。胡枝子多糖对·OH具有一定的清除能力,有明显的量效关系,最高·OH的清除率为69.1%。经过t检验的数据结果表明,胡枝子多糖具有抗凝血作用,能够显著延长凝血时间,且随着浓度的升高作用增强,最长凝血时间为26.73min。
Polysaccharide was obtained with water extraction method from leapedeza bicolor. The optimum condition of extraction was determined by the single factor experiment and the orthogonal experiment. The pharmacological activities of inhibition, antioxidant and anticoagulation about the polysaccharide of leapedeza bicolor were investigated. The results showed that the highest water extraction rate of polysaceharide from leapedeza bicolor could be obtained with a ratio of solid to liquid 1:20 (g/mL),100℃, 60 min, 2 times of extraction; and the highest extraction ratio was 3.75%. The polysaccharide of leapedeza bicolor had the strong inhibition activity. The test indicated that all the test samples had inhibition against E.coli, M. tetragenus. The scavenging ratio to ·OH was increased by a certain trend with the increasing the concentration of polysaccharide of leapedeza bicolor. The highest scavenging ratio to · OH was 69.1%. The t-test statistics showed that the polysaeeharide of leapedeza bicolor had high blood anticoagulation activity,which could significantly prolong the clotting time, and the power was stronger with the increasing of polysaecharide. The longest clotting time was 26.73 min.
