摘要
目的探索六味地黄丸3种入血成分马钱苷、芍药苷、丹皮酚对大鼠前脂肪细胞内cAMP水平的影响;通过肾上腺素诱导大鼠前脂肪细胞,探讨该方的作用机制。方法培养大鼠前脂肪细胞,在其生长旺盛时加入肾上腺素,分别加入含马钱苷、芍药苷、丹皮酚的培养液继续培养,以该细胞内的cAMP水平作为评价指标,以放射免疫法测定细胞内cAMP的含量。结果与空白组比较,马钱苷组差异有统计学意义(P<0.01),芍药苷及丹皮酚对细胞内cAMP水平无明显的作用。结论六味地黄丸主要入血成分之一马钱苷能降低大鼠前脂肪细胞内cAMP的含量,且可拮抗肾上腺素使大鼠前脂肪细胞内的cAMP水平升高的作用,并使其cAMP含量恢复到正常水平;马钱苷是六味地黄丸发挥药理作用的主要有效成分之一。
Objective To investigate the effects of loganin,peoniflorin,paeonol on the cAMP preadipocytes in rats and the mechanism of Liuwei Difang pills.Method The preadipocytes from rat were cultured in growth prosperity,the loganin,peoniflorin and paeonol were added in,then,the contents of cAMP in preadipocytes were determined with radio-immunity.Result Compared with blank group,the difference of cAMP in Loganin was distinctive(P 0.01),which suggested that Loganin was effective to lower the concentration of cAMP in preadipocytes,and depressed the epinephrine to increase the concentration of cAMP in preadipocytes,and the peoniflorin and the paeonol were not the same action.Conclusion Loganin can decrease the concentration of cAMP in preadipocytes,resist the epinephrine to increase the concentration of cAMP and restore the normal concentration,which means,loganin is one of the effective pharmacological constituents in the pills.
