摘要
目的探讨UPA、UPAR基因及蛋白表达与鼻咽癌的发生、发展及浸润转移之间的关系。方法应用原位杂交技术和流式细胞术分别检测鼻咽部慢性炎症患者20例、鼻咽癌无转移患者20例、鼻咽癌仅颈部淋巴结转移患者20例和鼻咽癌淋巴结及远处器官转移患者20例等4组患者组织中UPA、UPAR mRNA及蛋白的表达水平。结果各组中UPA、UPAR mRNA和UPA、UPAR蛋白质表达水平由高到低依次为:鼻咽癌远处器官转移组、鼻咽癌仅淋巴结转移组、鼻咽癌无转移组和慢性鼻咽炎症组。组间表达水平比较UPA mRNA、UPAR mRNA(P<0.05)和UPA、UPAR蛋白(P<0.01)均差异有显著性;在每组中UPA与UPAR蛋白表达水平之间均存在良好的相关性(P<0.01)。结论UPA和UPAR的基因和蛋白表达水平可随鼻咽癌的发生、发展、浸润转移而发生改变。UPA和UPAR的高表达在鼻咽癌浸润转移中起重要作用,是鼻咽癌预后不良的表现之一。
【Objective】To explore the relationship between the expression levels of UPA/UPAR mRNA and UPA/UPAR protein in the nasopharyngeal carcinoma tissues and the genesis,development,invasion and metastasis.【Methods】To detect the expression levels ofUPA/UPAR mRNA and UPA/UPAR protein in tissues by using in situ hybridization and flow cytometry system for the chronic nasopharyngitis group(20 cases),nasopharyngeal carcinoma without lymph node metastasis(20 cases),nasopharyngeal carcinoma only withlymph node(20 cases) and nasopharyngeal carcinoma with distal metastasis(20 cases).【Results】The expression levels of UPA/ UPAR mRNA and UPA/ U-PAR protein the nasopharyngeal carcinoma with distal metastasis,the nasopharyngeal carcinoma only with lymph node,the nasopharyngeal carcinoma without lymph node and the chronic nasopharyngitis(P0.05).There were statistical significance with comparison between the UPA protein and UPAR protein in each of two groups(P0.01).There was a significant statistical correlation between UPA protein and UPAR protein in each group(P0.01).【Conclusions】 The change of expression levels of UPA/UPAR mRNA and UPA/UPAR protein were found,which accompanying with the invasion and metastasis in nasopharyngeal carcinoma.Overexpression of UPA and UPAR may play an important role in the invasion and metastasis of the nasopharynaeal carcinoma,and show the poorest prognosis.
