摘要
目的:针对地衣初生、次生代谢产物丰富的特点,提出了一种适于地衣总DNA的提取方法。方法:应用改良的CTAB法,对3种常见地衣进行了总DNA提取,经过琼脂糖电泳、紫外吸收分析DNA质量。结果:3个样本DNA的A260/A280值均在1.8～2.0之间,琼脂糖电泳图像清晰。结论:改良的CTAB法简便、省时、价廉,适合于地衣总DNA的提取,获得的DNA含量高,纯度好。
Objective ： To select the best method to total DNA isolation from lichens which were rich in primary and secondary metabolites.Method ： The improved CTAB method was used to extract the total DNA from three common lichens.Extracted DNA was measured by agarose gel electrophoresis and spectrophometric analysis.Result： A260/A280 of total DNA from three sampled was 1.8 to 2.0,The agarose gel electrophoresis produced clear strips.Condusion ： Improved CTAB method which simple,save time and cheap,was good for extract high quality DNA from lichens.
