摘要
目的评价接种环粘取法血吸虫尾蚴检出效果。方法将血吸虫尾蚴分为10~100条8组,用接种环沿瓶壁在"品字形"3个部位,各取表面水膜3环。所取水膜移至载玻片,在显微镜下(40×)镜检尾蚴,计算尾蚴检出率。采用玻片法计数血吸虫尾蚴。结果 10、30和50条组尾蚴检出率分别为0、20.00%和40.00%;60~100条组尾蚴检出率为100%;每瓶粘取尾蚴数量平均为2~3.6条,环检出率在22.22%~31.11%。60条组与30条组比较尾蚴环检出率差异有统计学意义(χ2=8.3890,P〈0.01),与70~100条尾蚴组比较差异无统计学意义(2χ=1.7857,P〉0.05)。玻片法直接计数每只感染性钉螺尾蚴数最少为162条,平均(761±445)条。结论血吸虫尾蚴密度≥60条/250 ml时采用接种环粘取检查效果好,检出率为100%,适用于改良压碎逸蚴法取样。
Objective To evaluate the effects of using an adhesive inoculating loop to detect Schistosoma cercariae.Methods Schistosoma cercariae were divided into 8 groups with 10-100 cercariae.Cercariae were inoculated along the side of a bottle in 3 locations resembling a triangle.Aqueous film from the liquid surface at each site was sampled with 3 inoculating loops,and the samples were then placed on a glass slide under the microscope(40×).The rate of cercariae detection was calculated and the glass slide was used to count the number of cercariae.Results Groups with 10,30,or 50 cercariae has those cercariae detected at rates of 0,20%,and 40%.Groups with 60-100 had those cercariae detected at a rate of 100%.Each bottle with adhering cercariae had an average of 2 to 3.6 cercariae,loops helped to detect these cercariae at a rate of 22.22%-31.11%.There was a significant difference(χ2 =8.389 0,P0.01) in the rate at which loops helped to detect cercariae for the group with 60 cercariae and the group with 30 cercariae while there was no significant difference(χ2 =1.785 7,P0.05) in that rate for groups with 70-100 cercariae.Direct counting of the number of cercariae per infected snail using the glass slides indicated at least 162 cercariae,with an average of 761 ± 445 cercariae.Conclusion With a Schistosome cercariae density ≥ 60 cercariae / 250 ml,inoculating loops were effective at sampling,with a rate of cercariae detection of 100%.This can lead to improved sampling of crushed escaping larva.
