光谱法研究全氟癸酸与牛血清白蛋白的相互作用

摘要 采用光谱法研究了生理条件下全氟癸酸(PFDA)与牛血清白蛋白(BSA)的相互作用。结果表明,PF-DA借氢键和范德华力与BSA发生作用,并以静态方式猝灭BSA的荧光,其结合常数和结合位点数分别为2.9471×107L.mol-1和1.061。同步荧光和圆二色光谱数据说明,PFDA改变了BSA的二级结构。 The interaction of Perfluorodecanoic Acid(PFDA) with bovine serum albumin(BSA) was investigated using spectroscopic methods.The experimental results showed that PFDA quench the fluorescence of BSA with a static mechanism,in which hydrogen-bond and Van Der Waals force play a major role.The apparent binding constant/number of binding sites for PFDA-BSA were calculated to be 2.9471×107L·mol-1/1.061,respectively.Synchronous fluorescence and CD spectra confirmed that the secondary structure of BSA has been altered in the presence of PFDA.
